I have two partitions on a Storage Area Network that I need to use Partition Manager to resize one with unused space from the other, however when I try I get the following error message:

Partition Manager
Operation “Check file system” cannot be completed because volume G: is being used now.
Please try the following:
Manually close used files and try the operation
Confirm restart of your computer
Cancel the operation

On Paragon's web site there are comparisons of the different versions and it indicates that a hot resize can be performed without affecting windows or interupting it's work.
How can I troubleshoot this issue to resolution?

I have disabled all anti-virus services and processes and tried to manually resize the partitions, but still ge the same results when I click apply.
Does Partition Manager not play well with data stored on a SAN or should it work just as if it were a single server environment?

Hmm, I wouldn't use Partition Manager 8.5 to perform resize on NAS or SAN. Or take backup at least and only then perform resize. As far as I know this utility is designed to resize partitions on local HDDs only.

I tried this on a vista machine's local c drive and get the same message, so I am not sure that it is an issue with the SAN.
Any ideas on how to stop the volumes from being used in order to resize the partition, whether it is on a local hdd or a partition on a SAN?

Any assistance would be greatly appreciated.

In case you use the Redistribute Free Space Wizard then this message will come all the time because it makes check of file system start. Sure check of the file system must be performed in non Windows mode and that's why the tool from Paragon claims for files being opened. In other words, no application could be running from this partition or any file opened from it (it locks the partition). When you choose Restart Now the task will be performed in Windows start up mode. Like Windows CHKDSK when you use /f key.
I know that Paragon is going to release an update of Partition Manager that has better engine for Vista partitions, so I recommend you to wait a bit. It is promised to be released this week, let us see.
